The Hammers were without Nigerian defenders Olatunji Akinola and Ajibola Alese, while the Red Devils manager Richard Sbragia did not name Matthew Olosunde in his squad list after he suffered a serious injury against Derby County three weeks ago.
Manchester United conceded four goals despite twice leading in the first-half through Demi Mitchell's strikes.
A hat-trick from ex-Spain U17 star Toni Martínez and Joe Powell's goal on 65 minutes helped West Ham secure their fourth win in the division.
With United trailing by two goals, Tosin Kehinde came close to reducing the deficit on two occasions, one of which came back off the bar.
The Lagos-born midfielder had been nominated for Manchester United's Goal of the Month for September for his beautiful strike against Chelsea on September 18, an award picked up by Ecuador star Antonio Valencia.
